Estimation of Nicotine in Tobacco Extract Using C8 Column in High Performance Liquid Chromatography

Background: Nicotine is the most used toxic substance in spite of mass media awareness. Column estimation nicotine found to be inhibiting factor for its estimation.Objective: To develop a method on C8 column HPLC.Methods: The involved model Waters 1515 binary HPLC Pump system interfaced with 2489 UV/VIS detector, 4.6 X 250 mm, 5μm beads Symmetry at 37°C, and an isocratic mobile phase containing 60%: 40% v/ v mixture 10mM sodium acetate (pH 5.5) methanol flow rate 1.0ml/min 256 nm. was validated specificity, linearity, precision, accuracy, Limit Detection (LOD), Quantitation (LOQ) suitability.Results: peak average retention time 3.415 minutes observed chromatogram RSD <0.5%; linearity greater than 0.99 acceptable precision (within USP limits <2.0% RSD) tailing less 2. LOD 0.200 μg/ml 0.609 nicotine. yield extracted by acid base extraction from tobacco 1.68% (range: 1.34–2.22%).Conclusion: A based analysis developed successfully.International Journal Human Health Sciences Vol. 07 No. 02 April’23 Page: 166-170
